MatriClone is a new matrix designed to support iPSC growth in single seeding workflows while maintaining pluripotency. MatriClone is optimized for ‘in-solution’ seeding and well suited for automated imaging. When used as part of the VIPS PRO seeding protocol, MatriClone ensures high levels of single cell survival and subsequent outgrowth. This matrix simplifies the workflow by eliminating the need to coat plates. MatriClone is composed of a defined matrix containing a recombinant peptide and is animal component-free.

Our matrices can be used both in-solution, or to pre-coat plates. The term ‘in-solution’ refers to adding the matrix to the culture media at the same time as the cells during cell passaging, removing the need to pre-coat plates. The optimum coating density for both in-solution and pre-coating methods may differ depending on the cell line, application, and medium selected but don’t worry, our useful calculator is here to help! Select if you will be using in-solution or precoating, your surface area and number of plate to find out how much product of each matrix is required.

When it comes to using MatriClone or Clinical Manufacturing Grade laminin in-solution, the important thing to remember is that the required volume of matrix is independent of the well fill volume, it’s all about surface area.

For example:

  • To coat a 6-well plate (9.6 cm2/well) with 0.25 μg/cm2 following the in-solution method, 4.8 μL of MatriClone is required per well.
  • To coat all 6 wells in the plate, 28.8 μL of MatriClone should be added to 12 mL of cell suspension before seeding 2 mL of cell suspension, plus MatriClone, into each well.
  • Should the scientist decide to fill each well with 1.5 mL of medium instead, the quantity of MatriClone would still be 4.8 uL per well because the surface area of the well has not changed.

Supports Regulatory Approval

MatriClone and Clinical Manufacturing Grade Laminin are optically suited for whole well imaging, allowing scientists to capture crucial whole well images required for regulatory approval. The matrices, combined with the Cell Metric X and VIPS PRO technologies, enable extremely highest resolution images to be captured ready for submission.

Improved iPSC Adhesion

Laminin-511 is a major component of the basement membrane which is expressed in early development of the embryo. There are numerous forms of Laminin, all of which promote iPSC growth; however, this truncated version of the recombinant Laminin-511 promotes greater adhesion than full-length Laminin protein, in addition to other market alternatives.

Supports iPSC Pluripotency

MatriClone and Clinical Grade Laminin are comprised of the laminin 511 isoform which is instrumental to the growth and maintenance of iPSCs. Immunohistochemistry (IHC) staining has been used to confirmed the expression of important pluripotent markers: OCT4 and NANOG.

The term ‘in-solution’ refers to adding the matrix to the culture media at the same time as the cells during cell passaging. As MatriClone and Clinical Manufacturing Grade Laminin are soluble, there is the option to add it during plating rather than pre-coating the plates.

Using MatriClone/Clinical Manufacturing Grade Laminin in-solution saves time as you no longer need to coat your plates ahead. Simply add the matrix to your cell suspension and seed directly into a tissue culture treated plate!

Additionally, in-solution use requires much less matrix compared to pre-coating.

Please note that MatriClone/Clinical Manufacturing Grade Laminin must be used in-solution when seeding with Advanced Instrument’s VIPS PRO.

Yes, however this process is very simple. Once cells have reached confluence and are ready to be passaged, simply dissociate them following your standard protocol. Instead of plating the cells onto your traditional matrix, seed your cells into a plate which has been pre-coated with MatriClone/Clinical Manufacturing Grade Laminin, or add MatriClone/Clinical Manufacturing Grade Laminin during plating (if following the in-solution protocol).

Cells can be dissociated using any of the traditional dissociation reagents. Internally, we have experience with Accutase, TrypLE, Versene, ReLeaSR, GDCR and EDTA. We have found TrypLE and ReLeaSR to be the most popular amongst our customer base.

Yes, the matrices are suitable for clump and single cell passaging of iPSCs.
